The only comedy show I can think of which was done by brothers was The Smothers Brothers Show. I don't imagine that the Jacksons or Osmonds shows were considered comedy (more like variety), so I ruled them out.

As for the person, some guesses would be Sandy Duncan, Ann-Margret, Sammy Davis Jr, Gene Kelly, Dean Martin... and that's about it.
